First appearing in season four's "Minimal Loss (2008)", Benjamin Cyrus was loosely based, in part, on David Koresh, the target of the Waco siege in 1993. Having changed his name in 1990, and chosen as an allusion to the king of the first Persian empire, "Koresh" is Hebrew for "Cyrus".
In the beginning of the episode, each main character of the series is introduced as they were introducing themselves in the series.
While this is the third milestone episode of the series. This is the first time it's been a season premiere. While 100 and 200 were towards the middle of season 5 and 9 respectively.

On the plane ride to Kentucky, the team determines that Merva and his followers are taking Reid to a town called Arcadia. Joe Mantegna previously starred in Joan of Arcadia (2003).
