When the Bartender pours the whiskey for grant the ice was still solid, but after the cut the ice has melted considerably and the whiskey is diluted.
In the first scene with agent Coulson and Skye there is a drawing on the wall by Agent Coulson. The lowest circle on the right is only half complete, but in the next shot the circle is drawn in full.

Revealing mistakes

The way light reflects off Derek's tattoos reveals they are on the surface of his skin, not below it.
When Skye is on the phone with Grant, the phone's screen is on, illuminating her face, implying she's not really on the phone. The screen goes black when on a call.
(possibly intentional) When Grant Ward arrives at the Goldbrix Tavern, the sign on the door clearly reads Closed, but he walks right in and the bartender is behind the bar ready to serve drinks.

Plot holes

Grant shows Bobbi he is holding the bomb's dead man switch with his right thumb. After he gets up to retrieve his items, he uses both of his hands including his thumbs but the bomb does not explode.
