When Earl jumps out of his truck with his shotgun and orders Jacob to put Sissy down, Jacob has some blood on his upper back and the back of his head. After he kills Earl and picks up Sissy, there's no blood on his back nor the back of his head.
When Jacob kills Earl, he has more blood on his face and the front of his shirt. Then when he picks up Sissy, the amount of blood on his shirt, mostly on his left side has changed.
When Jacob arrives at the cemetery carrying Sissy, the blood stains all over him are different than they were after killing Earl.
When we first see the outside shot of Shepherd's Diner, the sign on the outside clearly reads "Shepherd's Diner." However, when we get inside the diner, the sings on the wall clearly read "Shepard's Diner."
