- The hijinks now move from the town of Great Falls to the University of Michigan, where the infamous Beta House fraternity has been setting new standards for incredible parties and outrageous antics. Our heroes are cousins Erik and Dwight Stifler. Erik has recently been dumped by his high school girlfriend and is trying to re-invent himself while pledging Dwight's fraternity, Beta House. Meanwhile, Dwight is trying to save Beta House from an onslaught by the newly established GEK ("Geek") House, which is led by power-hungry nerd leader Edgar and his band of geek brothers and sisters. Erik's best friend Mike "Cooze" Coozeman and his new roommate Bobby also pledge Beta House and ARE planning to enjoy college to the fullest. It promises to be a semester full of partying, craziness, pranks, and sexual tension to exceed expectations... ...and it does! Erik finds a girl, Ashley, to heal his heartbreak. Cooze dates Ashley's roommate Denise -- but finds himself in a possible "Crying Game" scenario. Dwight and the other Betas fight off Geek House for social supremacy on campus. The rivalry gets so intense that the only way to settle it is through direct competition in the Greek Games, an ancient campus ritual that went so far out of control it was banned forty years ago. It turns out that even Geeks can be bullies, so the Betas have to pull together to fend off the Geeks and prove that some traditions deserve to be upheld.—USHE

- After his girlfriend Tracy breaks up with him to be with her ex-boyfriend Trent, Erik Stifler (John White) and his friend Mike "Cooze" Coozeman (Jake Siegel) arrive at the University of Michigan as freshmen. Erik is dropped off by his father Harry (Christopher McDonald), who encourages him have some sex, to get Tracy off his mind. Harry shows Erik the list of girls he had sex with during his days. Harry mistakenly reveals he and his wife were separated for a week when Erik was 3 years old, and Harry had sex with 10 women in that week, including Cooze's mother.
Upon arriving at his dorm, Erik meets a girl named Ashley (Meghan Heffern) while she is taking a shower in the co-ed bathrooms. He then meets his new roommate Bobby (Nick Nicotera) and his girlfriend Margie while they're having sex. Bobby invites Erik to sit right next to him, while Margie was on top of him, riding him, completely naked.
Erik is then welcomed by his cousin, Dwight Stifler (Steve Talley), the leader of the Beta House fraternity. Dwight invites Erik, Cooze, and Bobby to a party. Erik and Cooze meet Ashley and her roommate Denise (Sarah Power) at the Beta House. Denise is a bit "prim and proper" and makes Ashley leave the rowdy party at Beta house to go to a party at the Geek house. Geek house used to be a social club till some rich kids came along, bought a fraternity house and set up a new charter. Now, one needs perfect SAT scores, needs to be filthy rich with a high future earnings potential, just to get in. They have billionaire alumni, and they run the student government, and the faculty loves them. They attract the hottest girls from the entire school as Geeks are going to be millionaires right out of school.
They meet Wesley / Blackout Menace (Jonathan Keltz), the rush chair of Beta House who experiences alcohol blackouts. He once adopted a Thai kid in Bangkok during one of his drunken binges and only realized it when he woke up on the plane ride back home. They learn how they can gain eligibility to pledge for the Beta House. They are also informed by Dwight that the Geek House, ran by wealthy nerds, has been trying to shut down Beta House. The Geeks believe that for a new civilization to prosper, the old one has to crumble.
Dwight tells Erik that college is his one chance to hit the reset button on life and Erik can choose to be anyone he wants to be. The party continues unabated at the Beta House.
Edgar (Tyrone Savage) head of the Geek house, lures away all the hot girls from the Beta house, by telling them their booze has run out, and the Geek house has champagne, a hot pool and swimsuits for everyone. Erik is drunk beyond control and barfs all over a hot girl, prompting all girls to move over to the VIP party at the Geek house. Dwight knows that Edgar is just throwing his money around but is powerless to stop him.
Later, Beta house votes in Cooze and Bobby, but are not sure about Erik since he vomited on a hot girl at the party and caused the party to move over to the Geek house. Plus, the house is livid that Erik passed on his chance to sleep with Brandy, whom they all consider to be the top of their list. Dwight pulls his rank at the house to get Erik a chance to get in. Dwight assigns the new pledges 50 tasks to complete before the end of the semester.
Erik and Ashley go on a first date at a restaurant where Erik has hot soup spilled accidentally on his legs, so Ashley brings him to her room to clean up. Ashley has Erik remove all his clothes and puts him in a towel. Ashley notices that Erik has a big red rash on his inner thigh.
As Ashley rubs lotion on Erik's thigh, he accidentally ejaculates on her belongings. Erik apologizes and explains to Ashley that, after breaking up with Tracy, he has not had an orgasm in four months due to masturbation being a taboo in his house because of a previous incident. Along with two other pledges, Erik, Cooze, and Bobby complete fifty tasks to pledge the Beta House, including: getting their behinds signed by a stripper with photo proof, having sex with a professor, marrying a fellow pledge, having sex in the Dean's office, drinking a gallon of milk without urinating, serenade a woman at the 901, do a traditional dance at the international, open an illegal casino, drive down to Mexico and get a tattoo, get extremely drunk and urinate in your pants, pose for the campus nude magazine, and placing a live ostrich in the Geek House. After they complete their final task stealing something from the Geek House. They steal a trophy motherboard from the Geek house, and Bobby takes an upper Decker dump in their bathroom.
Edgar, the president of the Geek House, challenges the Beta House to the "Greek Olympiad" which has been banned for 40 years on the college campus, with the winners taking the loser's House. Before the competition starts, Dwight learns that while attempting to rush the midget fraternity, Edgar had sex with a sheep.
Being the Olympiad's last winner, Beta House alumnus Noah Levenstein (Eugene Levy) returns to campus to officiate the competition. The first game is won by Beta House as Dwight and Edgar compete in removing girls' brassieres. Geek House win the next three matches: a boxing bout turned light-saber duel, catching a greased pig, and a two-person "69" race. Beta House wins the fifth and sixth competitions: a Russian roulette challenge between Dwight and Edgar with aged horse semen, and a beer drinking competition between all members of both Houses ultimately won by Wesley via a keg stand.
During the final lap dance challenge, Beta House uses Edgar's sheep fetish against him with Bobby's girlfriend Margie (Christine Barger) wearing a sheep costume, resulting in the Geek House losing the Olympiad.
Having won the Geek House mansion, the Beta House hosts a toga party. Erik and Ashley establish their relationship and have sex. Meanwhile, Cooze and Denise finally sleep together as well, and Cooze discovers that what Denise is hiding is the fact that she ejaculates during sex, much to his excitement. Wesley wakes up after a blackout and discovers that he had sex with the Geek House bodybuilder. Edgar instructs a stripper at the Silver Dollar to dance with the headpiece of the sheep costume.
