- High schooler Saya is a friendly amnesiac, who lives in Okinawa with her stepbrothers Kai and Riku and adoptive American father George, a retired Vietnam War vet. One night, mysterious bass violinist, Haji, saves Saya from a monster.
- [NOTE: Blood+ is the sequel series to the movie Blood: The Last Vampire (2000) in which Saya Otonashi is introduced as a young girl whose blood is the only known weapon against a race of blood-drinking beasts called the Chiropterrans. Following the events of Blood: The Last Vampire, which took place in 1966 Okinawa, Saya slipped into a deep coma. Her mentor David, a member of the Red Shield Organization, entrusted her care to his friend George Miyagusuku. Saya awakened from the coma a year ago and is now living in the Koza district of Okinawa with George and his two sons, Kai and Riku. They run a family restaurant/pub called Omoro. To Saya, they are her father and brothers albeit foster, because she cannot remember anything of her life prior to her awakening. She does experience short flashbacks, however, and feelings of familiarity, as when a U.S. bomber flies overhead. She needs periodic blood transfusions at a nearby clinic. David stops by occasionally to check on Saya's progress and to slip George some money for Saya's care. These summaries are based on the English-dubbed versions of the series. There appear to be some differences, particularly with names, between various versions, i.e., Haji/Hagi, Amshel/Anshel, Carl/Karl, Arjano/Argiano, Glay/Gray, Guy/Ghee.]
Tomorrow is Koza High School's track competition. After practicing on the high jump with her friend Kaori, Saya rides to the clinic with Kai on his scooter, only to find after she gets home that she left her track shoes at school. She assures her father that she'll be home in 30 minutes and heads back to school. She searches around the field, but her shoes are not to be found. Back at the family restaurant, Kaori has stopped by to drop off Saya's shoes, so George sends Kai to pick up Saya at the school. Suddenly, Saya is confronted by a tall, dark-haired man holding a dagger. He says, "Finally, we meet." Scared, Saya runs smackdab into Junichio Inomine, one of the teachers at the school. Mr Inomine checks around for the stranger, but he appears to have vanished.
Suddenly, Inomine is torn apart by a large monster. Saya runs into the school, but the monster breaks through a plate glass window and follows. Again, the tall, dark-haired stranger appears. He tosses his dagger at the monster, then picks up Saya and carries her upstairs into one of the labs. The stranger explains that the monster is a Chiropterran, a beast that lives on blood. He then cuts his own hand, which looks almost Chiropterran, and tries to get Saya to drink it, but she refuses. The Chiropterran pounces again, knocking Saya to the floor. The stranger jumps on Saya. Sucking his own blood, he kisses Saya and fills her mouth with the blood. "Saya, you must fight now," he says, just as Kai walks in.
