- The game begins with a narration by protagonist Jill Valentine. Within, she explains how the zombie infestation took control and went rampant through Raccoon City. With too few people fighting back the zombie hordes, the city became overrun.
On September 28. 1998. a full day before the events in "Resident Evil 2", it is revealed that the zombie outbreak appeared out of nowhere, and the zombies tore down and killed many people, assimilating them in the process. The police force and Umbrella soldiers attempted to stop the zombies, but were all killed and scattered. After a montage of death and destruction created in the zombies' wake, the title screen comes on, "Resident Evil 3." Former STARS member, Jill Valentine attempts to escape the city. She tries to convince a hiding man to escape with her but he refuses, heartbroken over the death of his daughter to the zombies. Most of the city's populace has been transformed into zombies due to an outbreak of the Umbrella's T-virus and Jill continues on alone, killing many of them who cross her path. Jill enters a bar where she sees STARS member Brad Vickers (helicopter pilot from the 1996 predecessor) being bitten and infected by a zombie. He tells her that a creature is after STARS members and that there is no escape. Jill continues and is attacked by infected dogs but makes short work of them with her shotgun. Upon leaving, she sees Brad being killed by the said mutation he was talking of earlier, the Nemesis program. She escapes and takes refuge in the police station.
She continues on her journey and meets three survivors, UBCS members Carlos Oliveira, Mikhail Victor, and Nicholai Zinoviev. Nicholai explains to Jill that they need to ring the bell of a clock tower and arrest the attention of the rescue team in helicopter patrol. Carlos is injured in battling zombies and both are nearly killed in a following explosion. Jill witnesses Nicholai's inhumanity in shooting an infected member of his crew while still conscious. Nicholai disappears and is presumed dead. The Nemesis Program attacks and corners Jill, Carlos, and Mikhail on a cable car headed to the tower. Mikhail sacrifices himself with a grenade which causes the car to crash into the courtyard and separate Jill and Carlos. Jill continues her way to the bell tower and arrests the attention of the helicopter but her plans for rescue are foiled when Nemesis arrives and blows apart the helicopter with a bazooka. The Nemesis then infects Jill with the T-virus before Jill defeats it, but then slips unconscious. Carlos arrives and brings Jill inside the clock tower to treat her infection. 3 days later, he cures her with a vaccination from the Hospital.
Upon awakening, Jill continues to Raccoon park where she enters the caretaker's cabin. There she finds Nicholai who reveals himself to be one of the "supervisors" who came to Raccoon City to gather combat data on Umbrella's Biological weapons, the Nemesis being targeted to kill all STARS members. Nicholai explains that the beast went out of control and killed Umbrella's troops as well, and that it was his job to record the live data of the beast's mission. Nicholai flees as Jill is attacked by a hideous membranous slug-like creature. After killing the creature, Jill escapes into an abandoned factory after being attacked by Nemesis, but throwing him off a bridge. Jill confronts Nicholai who plans to kill her and escape, having killed all the other supervisors to appear as the sole survivor and receive a "bigger bonus" as well as the reward for killing Jill. Just then, he is killed by an unseen creature and hung from the rafters. Jill is then attacked by Nicholai's creature, a further mutating Nemesis in the factory. Despite being heavily wounded by it, Jill manages to kill it by repeatedly shooting it with its own bazooka. Jill reunites with Carlos and the two board a rescue helicopter, taking off into the morning sky. They meet with Barry Burton who is revealed to be the pilot of the helicopter, and look down to see Raccoon city being blown away by a launched missile. "That's it," Jill states, "Umbrella's gone down."
The film ends on a News report explaining it was the president's decision to nuke the un-dead city, and commemorates the loss of lives in the city. The end credits show white text with black and white images of Raccoon city's structures before and after the missile's wake as well as the helicopter escaping.
