Joe's size varies immensely throughout film.
Max O'Hara challenges Mighty Joe Young with a tug of war against ten of the strongest men alive. Each man demonstrates a feat of strength as they are introduced with a different prop each is holding or wearing. Chains are ripped apart, crowbars and long steel bars are bent, a phone book ripped in half, a large wooden post broken, steel bands snapped off of bulging biceps. After the individual introductions are done the men are shown again as a group and you can see the props are all back to their original state. This is just a repeat of the previous shot of the group before they had a chance to mangle their props.
When the hobo tries to enter the moving van at the gas station the handle is on the left rear door and it opens first. When the cops open the doors a few moments later the handle is on the right door and it opens first.
When Jill throws Joe a banana at the beginning, she throws backhanded to her right, but the banana flies to her left and into Joe's right paw.
When Gregg starts to climb a rope up the orphanage wall, he places a table against the wall to step op on. In the next cut, the table is gone.

Revealing mistakes

At the end when Jill throws a banana to Joe, Joe catches the "banana" but the actual banana thrown can still be seen flying past Joe
When Joe pushes the truck free, his shadow is cast against the "distant" landscape, revealing it to be a backdrop.
When Joe makes his first appearance, and starts poking around the wagons containing the lions, watch the background carefully. Immediately to the left of Joe, between him and the far left wagon is a large bush. Just after Joe shoves the wagon the first time a light suddenly shines on the bush (presumably someone had repositioned a spotlight between shots) and from that point on you can see the reflection of Joe and the wagon reflected in what appears to be an unpainted area of the glass painting behind him.
When burning beams fall onto the police car, they pass 'through' the now transparent vehicle.

Crew or equipment visible

Max is sitting typing in his tent in Africa, a servant exits the tent, pulling back the net in front of the opening. For a brief moment a hand is visible pulling the net out of the shot.

Plot holes

At the Children's House, Jill never hears how many children are trapped inside. The woman only tells Gregg. Jill, however, runs in to save the two children. After, the two children are saved by Gregg. A third child appears unharmed at the very top floor of the house. This implies that the adults didn't count the children after evacuating.
