In the chase sequence at the end of the movie, Bailey damages the gate of the ticket booth (with the "OFFICE" sign) by ramming his arms through the bars, yet seconds later it is intact and undamaged.
Sebastian asks Dexter to be introduced to members of the city's baseball team "when we get to St. Louis." At this point the River Queen had already arrived in St. Louis.

Revealing mistakes

Crew members can be heard laughing during the "Who's On First?" routine (who could blame them?)
At one point in the "Who's On First" routine, Costello forgets Abbott's character's name (Dexter Broadhurst): he says, "Stay on third base, Misterrrrr . . ." and quickly glances off-camera while someone obviously mouthed or wrote the correct name, as Costello hurriedly says "Broadhurst".
As Bailey rushes through the door he has just stuck his head through, the door hits and disturbs the ceiling woodwork, revealing the flimsiness of the set.

Miscellaneous

20 seconds into Who's On First, Costello stumbles into the footlight and shatters it across the stage and improvs "Mr. Broadhurst, I didn't see the lights- I forgot about them." Abbott continues on, ignoring it after kicking away some broken glass.

Anachronisms

The movie is set in the 1890s, but several of the "period" songs played were written in the 1900s.
When the band plays and Sebastian goes down the wrong street with the drum, there's a 1940s car in the background.
The movie is set in the 1890s, but Life Savers candy (which is used as a joke in the movie) was first created in 1912 by Clarence Crane, a Cleveland chocolatier and father of the famed poet Hart Crane, who was looking for a new "summer candy" to supplement his chocolate business.
