- Robert Langdon is in Italy. He wakes up in a hospital with no memory of recent events. He appears to have suffered some head trauma. He's being treated by Dr. Sienna Brooks. Suddenly someone dressed in police uniform comes in and starts shooting, trying to get to Langdon. Brooks helps him. Langdon discovers he received an email from a friend in Italy who told him that he too is being hunted, and he's gone into hiding with what they took. Of course he has no idea what he is talking about. He finds something in his belongings that reveals the Map of Hell which is inspired by Dante's Inferno. He learns about a man Bertrand Zobrist, a billionaire who believes the world is too overpopulated and that drastic measures are needed. He plans to release a virus which he created to kill half the planet's population. - The geneticist Bertrand Zobrist is hunted down by three men and commits suicide from the top of a tower in Florence. Zobrist believes that the Earth's population that has an exponential growth should be reduced in 50% to save the other 50%. In Florence, Professor Robert Langdon awakens in a hospital with no memory of the last two days and with daydreams and dreadful visions. Dr. Sienna Brooks explains that he has a head trauma cause by a bullet that scratched his head and he would recover his memory in a few days. Out of the blue, a doctor tells that the police wants to interrogate him and a police officer, who is indeed the killer Vayentha, shoots the doctor but Sienna and Langdon succeeds to escape to her apartment. Langdon finds a Faraday pointer in the pocket of his jacket and when they project the image, they see the Map of Dante's Inferno by Sandro Botticelli. They contact the American Consulate but give the wrong address nearby the apartment. Soon the area is under siege of the World Health Organization (WHO) agents and the local police, including Vayentha, and Langdon and Sienna conclude that Zobrist has created a virus to annihilate the world's population. - Harvard University professor Robert Langdon (Tom Hanks) awakens in a hospital room in Florence, Italy, with no memory of what has transpired over the last few days but being plagued with visions of a scorched Earth. Dr. Sienna Brooks (Felicity Jones), one of the doctors tending to him, reveals that he is suffering from amnesia as a result of a bullet wound to the head. Another doctor says the police are there to question Langdon, but the officer turns out to be Vayentha (Ana Ularu), an assassin, who shoots the doctor while heading down the hall. Sienna helps Langdon to escape, and they flee to her apartment.
Among Langdon's personal belongings, Langdon and Sienna find a "Faraday pointer", a miniature image projector with a modified version of Sandro Botticelli's Map of Hell, which itself is based on Dante's Inferno. They soon realize this is the first clue in a trail left by Bertrand Zobrist (Ben Foster), a billionaire geneticist who believed that rigorous measures were necessary to reduce the Earth's growing population, and who committed suicide after being chased by armed government agents.
Langdon and Sienna figure out that Zobrist, who is obsessed with Dante, has created a virus he has dubbed "Inferno", with the potential of decimating the world's population. In the meantime, they have been traced by both Vayentha and the World Health Organization (WHO), who try to raid the apartment, forcing them to flee again. Sienna convinces Langdon that Zobrist left Langdon the first clue to the trail which leads to the virus, although Langdon does not remember ever meeting Zobrist. Langdon's knowledge of Dante's work and history, and of hidden passages in Florence, allows the two to follow clues such as letters and phrases which lead to various locations in Florence and Venice, while evading the assassin and WHO. Along the way, Langdon discovers that he stole and hid the Dante Death Mask, a crucial clue in the chain, an event he also has no memory. (it seems that Langdon has been on the trail once before and now has no memory of it).
While escaping, Langdon and Sienna are cornered by Vayentha, who warns Langdon that he was once "required", but now he was acting "stupid" prompting her bosses to get rid of him. But Sienna intervenes and ends up killing Vayentha.
Vayentha reports to her employer Harry Sims (Irrfan Khan), the CEO of a private security company called "The Consortium", who is acting on behalf of Zobrist. The WHO agents are headed by Elizabeth Sinskey (Sidse Babett Knudsen), an old lover of Langdon's, and are trying to prevent the release of the virus. Zobrist has provided Sims with a video message about the virus, to be broadcast after 24 hours of Zobrist's death. But Sims breaks him firm's protocols by watching the video and Shocked by its content, Sims allies with Sinskey to prevent the outbreak. Sims tells Sinskey that Zobrist had hired Sims organization to hide his work from WHO & Sinskey. All attempts by Sinskey to stop Zobrist had failed because of Sims.
Langdon uses the clues left by himself to trace the death mask of Dante in a Church in Florence. However, Langdon and Sienna are contacted by Christoph Bouchard (Omar Sy), a man purporting to be working for WHO, warning them that Sinskey has a double agenda and is after the Inferno virus for her own profit. The three cooperate for a while, until Langdon realizes that Bouchard is lying and seeking to profit from Inferno himself, forcing the duo to flee on their own again.
Langdon figures out that the virus is in the Hagia Sophia in Istanbul. With that knowledge, Sienna abandons Langdon, revealing that she was Zobrist's lover and that she will ensure the release of the virus. Zobrist and Sienna used to play treasure hunt games; this trail was the backup plan in case something happened to Zobrist. Langdon is recaptured by Bouchard, but is rescued by Sims and re-teams with Sinskey, who asked him for help in interpreting the imagery from the Faraday pointer. Sims reveals he was hired by Sienna to kidnap Langdon when Zobrist had been killed and drugged with Benzodiazepine to induce a memory loss; the events in the hospital were all staged. Sims broke ranks from Sienna when he realized what he was up to.
They realize the virus is in a plastic bag hidden under water in the Basilica Cistern in Istanbul. The WHO team-along with Langdon, Sims, and Sinskey-race to locate and secure the bag, while Sienna and her allies attempt to detonate an explosive that will rupture the bag and Aerosolize the virus. Sims is killed by Sienna, and when Langdon confronts her, she commits suicide in an attempt to release the virus, but the bag is secured in time and Sienna's enforcers are killed. The virus is then taken by WHO, and Langdon goes back to Florence in order to return the Dante Death Mask.
