No. This movie is not connected to the "Timm-verse" of Batman/Superman/Justice League shows; although, Bruce Timm is art directing the project. "Style-wise, the film will feature an all-new look from animation guru Bruce Timm," according to Sander Schwartz, president of Warner Brothers Animation. The story is pretty streamlined due to the 75-minute running time. The role of the rest of the Justice League will be eliminated. The "World Without a Superman" segment makes up the second act of the movie. The "Reign of the Supermen" arc is replaced with a plot by Lex Luthor which uses some of the same beats, but with a very different twist. Edit (Coming Soon)
The first casting announcements were made on Saturday, March 3rd, 2007 at WonderCon in San Francisco, California. Adam Baldwin voices Superman/Clark Kent; Anne Heche is Lois Lane; and, James Marsters plays Lex Luthor. - Requiem and Rebirth: Superman Lives! A retrospective look at the craze which caught the world by storm. What happens when millions of fans are suddenly subjected to the death of their favorite character? Total mayhem! Going on record about the Death of Superman will be Paul Levitz and other DC staff as well as comic book artists throughout the industry. News clippings, reports and first-hand archival testimony from fans will be pieced together in a retrospective way. The featurette will also include a closer look at the dark Superman appearing in The Return of Superman graphic novel.

No, as the movie takes a different path than the comics. However, a new character, Lex's Superman clone, seems to combine elements of two of the previous Superman replacements (the "Superboy" clone and the Eradicator) and he also served in a general villain role similar to Cyborg Superman in the comics.
