Fazio-Londe disease (FLD), also known as progressive bulbar palsy of childhood, is an extremely rare genetic motor neuron disease affecting children and teenagers, it involves progressive paralysis of muscles due to degradation of the cranial nerves. PLD is caused by a mutation of the gene that produces riboflavin (vitamin B2), riboflavin being essential for cell energy metabolism, without sufficient levels of riboflavin motor neuron cells are unable to be regenerated. PLD causes rapid progressive weakness of tongue, face and pharyngeal (part of the throat behind the mouth and nasal cavity) muscles. It then progresses to paralysis of the facial muscles, loss of hearing and soon after paralysis of the muscles in the diaphragm. The mean onset age is eight years, progression to death can take as little as nine months, in other cases it can take years. There is no known cure, however progression of symptoms can be slowed with high does of riboflavin-5-phosphate (a metabolite of riboflavin), in some children it can even prevent progression to the fatal stage.
