The Playboy publishing mogul, 84, proposed to Playmate Crystal Harris on Christmas Day and was due to tie the knot with the 24 year old on Saturday.
However, TMZ.com reports suggested Hefner's bride-to-be had called off the nuptials and moved out of the publisher's Hollywood mansion - five days before the big day.
And Hefner has now taken to Twitter.com to announce the news in the same way he announced the shock engagement.
He writes, "The wedding is off. Crystal has had a change of heart."
Harris' manager Michael Blakey tells People.com, "The split was a mutual decision and the two remain good friends."
"I'd like to see Hef settle down. I just want it to be with the right person, and I feel like he's making a hasty decision. I'm not sure Crystal is the best thing for him," she told Life & Style magazine. "Crystal hasn't been around very long - she's a mysterious character. No one knows much about her. Plus, she's very young, probably too young to settle down."
Holly, 31, also revealed she is worried aspiring singer Crystal is using Hugh to start her career and fears he could end up losing everything in an expensive divorce.
